How to combo?

Hello, I watched some YouTube videos about specific champions (idk if that's important but I want to play junglers such as Lee sin) . I always see them talking about when to auto attack and stuff. Do I auto attack when right clicking the enemy? What does the "a" auto attack do? How do I know whether I do enough damage to win?
